Halo. A war that scale has got to have tons of great stories.
Axis & Allies: Iron Blitz - the game failed because of a weak AI, but it had the concept of mixing up the starting countries for each player. So by extention we'd be talking about an alternate universe WWII where anything goes. I'd like it in a mild dieselpunk format, where the altered war can gradually design devices never seen in our world.
Sid Meier's Alpha Centauri colonization space saga.
Freelancer. Compelling story, big universe. Enough similarities that it would probably interest the Firefly Browncoat fans.
Simply the best space sim I've ever played. It had a great story about megacorporations vs colonists on the fringes of known space. It had crazy pirates, cool ships, epic space battles, the lot. AND it had Bruce Campbell!
Would make a great TV show. In the game the player chooses to side with either the megacorporation or the colonists. You could do both in the show. Like every other episode is from one perspective or the other. Could offer some interesting opportunities, like seeing some of the same battles or events from both sides.
Could even get The Chin himself to star in it... assuming that Burn Notice will ever end.

1. Mass Effect as a TV show. I think it could hold it's own as a weekly TV show, and it would be most excellent for viewing on a Friday night.
Mass effect would be a brilliant tv series set over 4 or 5 years. The game is all ready very cinematic to begin with so it should work as a tv show. It always reminded me of babylon 5 as I played it, and i loved that show.
