That is a tough one, Chuck Norris is hands down the best in martial arts, but it took several nuclear blasts by military testing to kill John Wayne, and even then it took several year there after to do the job. So I would have to say The Duke

Ok Based on the parts they played as an actor, who in your mind is the tougher guy?
Is it:
A. John Wayne
or
B Chuck Norris

The question is an impossibility to determine, as such is a red herring.
Chuck Norris has too much respect for John Wayne to even try to be as tough as the Duke. That's why Chuck Norris decided to come to Earth in a human form after John Waynes' prime. Even when John Wayne had cancer he still was kicking butt and making interns take names.
OIC in the movies...hmm, well Chuck Norris did grab and stop a chainsaw in a movie once, so I guess him.

John Wayne. There is none tougher than The Duke. I give Chuck his propts, and I agree that it may well be that Chuck, out of sheer respect, would not outdo The Duke, The Duke was and is the most awesome, and had some of the best lines.
